Horseradish peroxidase, a common marker enzyme for antibodies, can catalyse the chemiluminescent oxidation of luminol in the presence of hydrogen peroxide. This transformation is used in ultra-sensitive ELISA assays. Luminescence can be detected at very low levels using extremely sensitive scintillation counters. Luminol is also used for the detection of blood stains in criminal investigation. For this luminol is mixed with a liquid containing hydrogen peroxide. If the mixture comes in contact with hemoglobin, the iron in the hemoglobin accelerates chemiluminescent oxidation of luminol.
Store at 12-18°C. Keep under Inert Gas.
Preparation of Luminol Stock Solution
and Working Solution in PBS
Stock Solution
1. Dissolve 17.7 mg of luminol (Gold Biotechnology, Catalog No. LUM)
in 10 mL of of DMSO.
2. Prepare 2 mL aliquots and store at -20°C and protect from light
3. The final concentration is 10 mM
Working Solution
1. When ready to use luminol solution remove from freezer to thaw
2. Add 250 μL of 10 mM stock solution to 10 mL of PBS (final
concentration of 250 μM)
3. Protect from light and discard any unused portion

Stock Solution

- Add 250 μL of 10 mM stock solution to 10 mL of phosphate buffered saline (PBS) solution with a final concentration of 250 μM
- Protect from light
